    • Bioavailability and disposition of solanine in rats and hamsters

      The toxicokinetics of [3H]-alpha-solanine after oral (po) and intravenous (iv) administration in rats and hamsters were studied, in order to decide which is the most appropriate model in risk assessment studies. The iv dose was 54 mug/kg; the oral dose was 170 mug/kg. After iv administration, the toxicokinetics of total radioactivity in blood were comparable in rats and hamsters. However, the clearance of total radioactivity from plasma was more effective in rats than in hamsters The half-lives of distribution and of the terminal phase of unchanged alpha-solanine were not different between rats and hamsters, whereas the systemic and the metabolic clearance were about 1.6 and 2.7 times higher in rats than in hamsters, respectively. The clearance of unchanged alpha-solanine is more effective than that of total radioactivity. After oral administration in rats and hamsters, the mean bioavailability of total radioactivity is about 29% and 57%, respectively, whereas the bioavailability of unchanged alpha-solanine is only 1.6% and 3.2%, respectively, when compared with iv administration. T1/2 el of alpha-solanine after oral administration was in rats a factor of four and in hamsters a factor of two shorter than after iv administration. A strong retention of radioactivity was seen in the hamsters after oral administration; only 40% of the dose was excreted within seven days vs 90% in rats. In some hamsters, severe damage of the duodenal wall was observed after oral administration. Based on these data, it was decided that the hamster is a more appropriate model in (sub)-chronic toxicity studies with alpha-solanine than the rat.<br>

Gegevens over chemische structuren, detectie methoden van ciguatoxinen (CTX), bronnen van CTX, marine organismen die worden geassocieerd met CTX, de toxiciteit van CTX in mens en dier, mogelijke preventieve maatregelen tegen ciguatera vergiftiging, rapportage van CTX-vergiftigingsgevallen, regulering en monitoring van CTX zijn beschreven. Tenslotte worden nog een aantal aanbevelingen gegeven voor een betere controle van het potentiele CTX-probleem in de toekomst.

      This review on ciguatera fish poisoning contains information on the ciguatera intoxication syndrome and the provoking ciguatoxins (CTXs) and gambiertoxin-4b (GTX-4B), of which CTX-1 is a major component at the end of food chain (the carnivore fish). Data on chemical structures and detection methods of ciguatoxin (CTX), sources for CTX, marine organisms associated with CTX, toxicity of CTX for animals and man, possible preventive measures for ciguatera intoxication, case reports of outbreaks of ciguatera intoxication and regulations and monitoring of CTX are included. Finally some recommendations are given for a better control of the putative CTX problem in the future.

    • Evaluation of the health risk resulting from exposure to polycyclic aromatic hydrocarbons in coal-tar shampoo

      Because several PAHs, among which benzo(a)pyrene (BaP), are considered to be carcinogenic substances, the safety of these products has been questioned. An estimate of external dermal exposure to BaP has been generated by application of a mathematical model. This estimate was the starting point in an assessment of the additional risk on local skin tumours. In this assessment risk estimates were derived for two different contact times and for two different periods of use. In the most optimistic scenario a concentration of about 1000 mug BaP/kg product results in an additional risk on dermal tumours of 1 per 10 exp.6 exposed persons. In the most pessimistic scenario, the same risk level (1 per 10 exp.6) is reached at about 100 mug BaP/kg product. Because of several uncertainties, the risk of the entire mixture of PAHs in coal-tar shampoos cannot be assessed. In order to compensate for these uncertainties, an additional factor of 10 can be applied to the concentrations of BaP which are associated with the accepted levels of additional risk. Thus, in the most optimistic scenario a maximal concentration of 100 mug BaP as indicator for total PAH/kg product would be advisable, while for the most pessimistic scenario a maximal concentration of 10 mug BaP as indicator for total PAH/kg product would be advisable. Therefore, the estimates of concentrations for total PAH in coal-tar shampoos which are associated with the accepted risk level of 1 per 10 exp.6 exposed persons may implicate an underestimation of the risk on systemic tumours.
    • Evaluation of the health risk resulting from exposure to polycyclic aromatic hydrocarbons in coal-tar shampoo

      Omdat verscheidene PAK, waaronder benzo(a)pyrene (BaP) worden beschouwd als carcinogene stoffen, zijn er vraagtekens gesteld bij de veiligheid van deze producten. Een schatting van de externe dermale blootstelling aan BaP is afgeleid door toepassing van een mathematisch model. Deze schatting was het uitgangspunt in een beoordeling van het additionele risico op huidtumoren. Er werden risicoschattingen afgeleid voor twee verschillende contacttijden en voor twee perioden waarover de shampoo wordt gebruikt. In het meest optimistische scenario resulteert een concentratie van ongeveer 1000 mug BaP/kg product in een additioneel risico op huidtumoren van 1 per 10 exp.6 blootgestelden en in het meest pessimistische scenario wordt dit risiconiveau (1 per 10 exp.6) bereikt bij een concentratie van 100 mug BaP/kg product. In verband met een aantal onzekerheden kan het kankerrisico van het gehele mengsel van PAK in koolteershampoo niet precies worden vastgesteld. Om voor deze onzekerheden te compenseren kan een extra factor van 10 worden toegepast op de concentratie van BaP die samenhangt met het geaccepteerde niveau van additioneel risico. Uitgaande van het meest optimistische scenario zou een maximale concentratie van 100 mug BaP als indicator voor totaal PAK/kg product raadzaam zijn, terwijl in het meest pessimistische scenario een maximale concentratie van 10 mug BaP als indicator voor totaal PAK/kg product raadzaam zou zijn. De schatting van de concentraties aan totaal PAK die een acceptabel risico inhouden op huidtumoren kunnen een onderschatting betekenen voor het risico op systemische tumoren.<br>

    • Guidance document on the derivation of maximum permissible risk levels for human intake of soil contaminants

      This report contains a basic step-to-step description of the procedure followed in the derivation of the human-toxicological Maximum Permissible Risk (MPR ; in Dutch: Maximum Toelaatbaar Risico, MTR) for soil contaminants. In recent years this method has been applied for a large number of compounds (the results have been published as separate RIVM reports). In the report the different steps in the procedure are discussed briefly and an outline of the kind of problems to be dealt with, is provided. General issues of regulatory risk assessment aimed at the derivation of health-based limit values are addressed and reference is made to some basic background literature. Overall, the approach for MPR-derivation as described here, is a pragmatic one in that use is made of existing toxicological evaluations by national and international bodies. Thus, it is attempted to avoid unwanted duplication of effort. In case no adequate review was available for a particular compound, a complete literature search for original publications is done. The results, however, were used selectively. The MPR-derivation as a whole proceeds in a structured manner, it is nevertheless stressed throughout the report that, unavoidably, for an adequate evaluation of the data on the different toxicological endpoints some professional judgement is needed.
    • Guidance document on the derivation of maximum permissible risk levels for human intake of soil contaminants

      Dit rapport bevat een basale beschrijving van de achtereenvolgende stappen in de gevolgde procedure bij de afleiding van het humaan-toxicologische Maximum Toelaatbaar Risico (MTR) voor bodemcontaminanten. In de afgelopen jaren is deze methode toegepast voor een groot aantal chemische stoffen (deze resultaten zijn gepubliceerd als aparte RIVM-rapporten). Tevens wordt een omschrijving gegeven van de problemen die zich bij de stappen kunnen voordoen. Op de meer algemene aspecten van de risicoschatting, gericht op het afleiden van grenswaarden voor chemische stoffen, wordt ingegaan en verwijzingen naar enige elementaire achtergrondliteratuur worden gegeven. De aanpak bij de afleiding van MTR's zoals hier beschreven is pragmatisch omdat gebruik werd gemaakt van reeds bestaande toxicologische evaluaties zoals opgesteld door nationale en internationale instanties. Op deze manier werd gepoogd ongewenste duplicering van werk te vermijden. Aanvullend literatuuronderzoek is zo selectief mogelijk verwerkt. De afleiding van de MTR zoals hier beschreven verloopt via een vastliggende werkwijze maar niettemin is het een onvermijdelijk en regelmatig in het rapport terugkerend punt dat voor een adequate beoordeling van de verschillende toxicologische eindpunten enige vakinhoudelijke expertise nodig is.<br>
    • De HDL-cholesterolbepaling en de veranderingen in het lipoproteinpatroon bij de rat onder verschillende dieet-omstandigheden

      Doel van het onderzoek was het ontwikkelen van een snelle betrouwbare micro-HDL-cholesterolbepaling in serum van de rat die toepasbaar was onder verschillende dieet omstandigheden. Drie groepen van 10 mannelijke Wistar ratten (RIV-Tox) kregen respectievelijk een semisynthetisch dieet (SSP-TOX), eenzelfde dieet met 1% en met 2% cholesterol aangeboden. Na 7 maanden werd in gepoolde sera van de verschillende groepen de cholesterolconcentraties in de verschillende lipoproteinen fracties bepaald m.b.v. ultracentrifuge. De HDL-cholesterolconcentraties bedroeg in de verschillende sera resp. 1,632, 0,849 en 0,815 mM. Naast deze verlaging van de HDL-cholesterolconcentratie was er een geringe toename van de totale cholesterolconcentratie. Deze toename werd vooral veroorzaakt door een toename van alle lipoproteinen met een dichtheid <1,040 g/ml. De uitkomst van de HDL-cholesterolconcentratie werd vergeleken met de uitkomst van de HDL-cholesterolconcentratie na lipoproteinprecipitatie met de combinatie fosforwolfraamzuur en magnesiumionen en de combinatie heparine met mangaanionen. De combinatie heparine 0,1% w/v en 50 mM Mn 2+ kwam het beste overeen met de resultaten van de ultracentrifuge.<br>
    • Health Risk Assessment for Organotins in Textiles

      In January 1998 RIVM was asked to carry out a preliminary risk assessment on organic tin compounds (organotins) in textiles. Measurements carried out by the Dutch Health Protection Inspectorate had shown these potentially toxic compounds to be present in several consumer products, including items of clothing worn by adults and children and hygiene products for women and infants. The initial assessment of this problem by RIVM (dated March 1998) was used in the ongoing discussions between the Health Indpectorate and industry. At an early stage in these discussions the industry indicated the sources of organotin contamination to heve been located and subsequently removed from the production process, thus reducing the issue under investigation to the question of whether there had been a risk for consumers at the time when the contamination was still in place (historical risk). In addition, industry provided further information that could be used to improve on the initial risk assessment. These new data largely cleared up the points of concern noted in our preliminary risk assessment. Some data limitations remain but overall the conclusion is that most likely the contaminated items did not pos any health risk for the consumers using them.
    • Health Risk Assessment for Organotins in Textiles

      In januari 1998 werd het RIVM gevraagd een voorlopige risicobeoordeling uit te voeren voor organische tinverbindingen (organotins) in textiel. Metingen uitgevoerd door de Inspectie Gezondheidsbescherming, Waren en Veterinaire Zaken wezen op de aanwezigheid van deze potentieel toxische verbindingen in diverse consumentenproducten waaronder kleding voor volwassenen en kinderen en producten voor hygienische verzorging van vrouwen en jonge kinderen. De eerste beoordeling van dit probleem verscheen in maart 1998; dit advies werd ingebracht in het lopende overleg tussen de Inspectie en de industrie over dit probleem. Reeds in een vroege fase gaf de industrie aan dat de bronnen van verontreiniging waren opgespoord en vervolgends uit het productieproces verwijderd. Daarmee was de vraagstelling teruggebracht tot de beoordeling of er sprake was van een risico in de periode dat de verontreiniging nog wel aanwezig was (historisch risico). Bovendien leverde de industrie verdere informatie die gebruikt kon worden om initiele risicobeoordeling nader uit te werken. Deze nieuwe gegevens gaven grotendeel antwoord op de punten die in het voorlopige advies wezen op mogelijke risico's voor de gezondheid. op enkele punten blijven er beperkingen in de beschikbare gegevens maar overall concluderen we dat de verontreinigde items waarschijnlijk geen enkel risico vormden voor de gezondheid van de consument.<br>
    • Immunotoxiciteit van natuurlijke toxinen. Een literatuur overzicht

      The data about the possible toxicity of natural toxins indicate that many of these toxins are immunotoxic. This was found in various animal species and in man. The effects on the immune system often involve clinical implications, i.e. reduced resistance towards infections. For many of the natural toxins, however, investigations on the immunotoxic effects are far from complete. The relation of the observed effects on the immune system to other toxic effects is often not clear. On several occasions effects on one or more immunological parameters have been described, whereas the significance for the ultimate functionality remains unclear. This means that the knowledge about the potential immunotoxicity is fragmentary for the majority of the natural toxins. The knowledge on the toxicological profile of many less well investigated natural toxins would be desirable. In particular attention should be given to the immune system as a possible target for toxicity. The mycotoxins Fumonisin B1 and Ochratoxin A should be considered for further immune toxicity studies, in view of the toxicological significance, the gaps in the knowledge and the possibilities to have enough material available for (immune-)toxicity studies.<br>
