Om de inschatting van de mate van ongerief
onafhankelijk te maken van ervaring en interpretatie van de persoon die de
observatie uitvoert, maken wij in onze studies gebruik van een volledig
geautomatiseerde opstelling. Met deze geautomatiseerde opstelling kan de
bewegingsactiviteit van individuele muizen 24 uur per dag gemeten worden.
Met name de nachtactiviteit is zeer gevoelig voor gedragsveranderingen,
waardoor deze parameter geschikt blijkt voor de inschatting van de mate van
ongerief. Om de reproduceerbaarheid van het model aan te tonen, wordt in
dit rapport de nachtactiviteit van vrouwelijke Balb/c/Rivm muizen die
behandeld zijn met PBS of Freund's compleet adjuvants (FCA, Difco
Laboratories, Detroit Michigan, USA) over de afgelopen jaren vergeleken.
Uit de resulaten blijkt, dat het verloop van de nachtactiviteit van groepen
muizen die dezelfde behandeling hebben gehad (PBS of FCA s.c. en i.p.),
exact hetzelfde is over de jaren heen.Geconcludeerd kan worden dat de
nachtactiviteit zoals wij die in ons model meten, in hoge mate
reproduceerbaar is.
We used a fully automatic system based on the locomotor activity of individual mice over 24 h to assess the degree of distress in female Balb/c/Rivm mice. In this way the assessment of distress could be ascertained independent of the experience and personal interpretation of the observer. The nocturnal activity was found particularly susceptible to changes in the normal behaviour of the animals, which can be used as an expression of distress. The nocturnal activity of these female Balb/c/Rivm mice treated with PBS or Freund's complete adjuvant (FCA - Difco Laboratories, Detroit MI, USA) was compared over the last five years to demonstrate the reproducibility of the model. The results showed the course of the nocturnal activity to be exactly the same for groups of animals with the same treatment using PBS or FCA. In conclusion, measuring undisturbed nocturnal activity is to a large extent reproducible.
