Een van de taken van het Communautair Referentie Laboratorium voor Salmonella (CRL-Salmonella) is het verbeteren van detectie methoden voor Salmonella. Hiertoe werd onderzoek uitgevoerd om de isolatie van Salmonella uit kippenfaeces te verbeteren. In vergelijking met de standaard ISO methode werden de volgende effecten bestudeerd: (i) de invloed van de incubatietijd van de voorophoping in gebufferd pepton water (BPw); (ii) het gebruik van Modified Semi-Solid Rappaport Vassiliadis Medium Base (MSRV) als selectief ophopingsmedium na voorophoping in BPw; en (iii) de invloed van direct selectief ophopen in Rappaport-Vassiliades (RV) en Rappaport- Vassiliadis Soya Pepton (RVS). De beste resultaten werden verkregen met gebruik van MSRV beent na 4 of 7 uur voorophoping in BPw. Goede resultaten werden tevens verkregen door direct selectieve ophoping van de faeces in RV of RVS, maar deze procedure was arbeidsintensiever dan de MSRV methode.<br>One of the tasks of the European Community Reference Laboratory for Salmonella (CRL-Salmonella) is the improvement of methods for the detection of Salmonella. Therefore, research was done to improve the isolation of Salmonella from chicken faeces. The following effects were studied in comparison to the standard ISO method:(i) the effect of incubation time of the pre-enrichment in BPw (Buffered Peptone water); (ii) the use of Modified Semi-Solid Rappaport Vassiliadis medium (MSRV) as selective enrichment medium after pre-enrichment in BPw; and (iii) the effect of direct selective enrichment in Rappaport-Vassiliadis (RV) and Rappaport-Vassiliadis Soya peptone (RVS) medium. The best results were obtained with the use of MSRV inoculated after 4 or 7 hours pre-enrichment in BPw. Also direct selective enrichment of the faeces in RV or RVS yielded good results but this procedure was more labor intensive than the MSRV method.<br>
