    Dit rapport beschrijft de resultaten van een in vitro experiment naar het methemoglobine inducerende vermogen van nitriet in humaan bloed. Er werd gebruik gemaakt van verse, gehepariniseerde, veneuze bloedmonsters. Tien natriumnitriet doseringen, in een molaire verhouding, nitriet : hemoglobine, tussen 0.01 en 0.15, werden getest. Elk experiment werd in triplo uitgevoerd. De bloedmonsters werden op een temperatuur van 37 graden Celsius gehouden. De hemoglobineconcentratie, het percentage methemoglobine en de pH van het bloed werden gedurende 2 uur op vaste tijdstippen gemeten. Geen van de bloedmonsters toonde zichtbare hemolyse van het plasma na afloop van het experiment. Er werden methemoglobine percentages tot 24% geinduceerd. Er werd een lineaire relatie gevonden tussen de natriumnitriet doses en het maximale methemoglobine percentage dat werd bereikt. De lineaire regressie analyse toonde een gecorrigeerde R exp.2 > 0.99 (p=0.0001). De verhouding, natriumnitriet doses : maximaal geinduceerde hoeveelheid methemogobine in het bloedmonster, was ongeveer 0.7 voor elke dosering.<br>
    This report describes the results of an in vitro experiment, investigating the methemoglobin inducing potential of nitrite in human blood. Fresh, heparinized, venous blood samples were used for the experiment. Ten sodium nitrite doses, with a molar ratio of nitrite to hemoglobin ranging between 0.01 and 0.15, were studied. Each sodium nitrite dose was tested in triplicate. During the experiment the blood was maintained at 37 degrees Celsius. The hemoglobin concentration, the percentage of methemoglobin and the pH of the blood were determined at timed intervals, for two hours following the start of the experiment. None of the blood samples showed visible hemolysis of the plasma after completing the experiment. Methemoglobin levels up to 24% were induced. A linear relationship was established between the nitrite dose and the maximum methemoglobin level that was induced in the blood. Linear regression analyses showed an adjusted R exp. 2 of > 0.99 (p=0.0001). The molar ratio of the nitrite dose to the maximum amount of methemoglobin that was induced at t-max, was approximately 0.7 at all dose levels.<br>
