    Als onderdeel van het EU-project 'Bacteriofagen in zwemwater' werd onderzoek uitgevoerd ter optimalisatie van de methode voor de detectie en enumeratie van somatische colifagen in water, zoals beschreven in ISO/CD 10705-2 van augustus 1995. Geconcludeerd werd dat deze concept ISO aangepast dient te worden op een aantal onderdelen. Het betreft onder andere de volgende aanpassingen. Voor het bepalen van het kiemgetal van de gastheer cultuur WG Escherichia coli dient membraanfiltratie of de gietplaat methode gebruikt te worden in plaats van de spreidplaat methode, daar deze laatste voorkeur dient een vers gekweekte inoculum cultuur van WG5 en geen ingevroren inoculum cultuur geruikt te worden, daar invriezen van de inoculum cultuur de faagtellingen negatief beinvloedt. Voor de faagbepaling heeft de Double Agar Layer methode (DAL) de voorkeur boven de Single Agar Layer methode (SAL), daar de DAL methode hogere resultaten heeft dan de SAL methode.<br>
    As part of the EU-project 'Bacteriophages in bathing water' research was carried out to optimise the method for detection and enumeration of somatic coliphages in water, as described in ISO/CD 10705-2 of August 1995. It was concluded that this draft ISO standard needs to be amended for certain aspects. This concerns among others the following aspects. For determining the viable count of the host culture WG5 Escherichia coli membrane filtration or pour plate technique should be used instead of spread plate technique, as the latter gives lover and less reproducible results than the other two techniques. A freshly prepared inoculum culture is found to negatively influence the phage counts. The Double Agar Layer method (DAL) is preferred to the Single Agar Layer method (SAL) for performing the phage analysis, as the DAL method gives higher phage counts than the SAL method.<br>

    Export search results
