dc.description.abstractDit rapport bevat de lezingen, conclusies en aanbevelingen van het seminar "Performing aquatic toxicity tests with poorly soluble substances", gehouden op 17 maart 1995 te Bilthoven. De grote internationale belangstelling voor het seminar gaf aan dat er dringend behoefte is aan richtlijnen en afstemming tussen regulerende instanties, industrie en contractlaboratoria bij de classificatie en risicobeoordeling van chemicalien die slecht in water oplosbaar zijn. De problemen doen zich met name voor bij de uitvoering van aquatische toxiciteitsexperimenten en bij de interpretatie van de resultaten van deze experimenten, omdat huidige richtlijnen niet blijken te voldoen voor deze stoffen. Doel van het seminar was het uitwisselen van informatie over de technische aspecten van de uitvoering van ecotoxicologische testen met slecht in water oplosbare stoffen en over problemen die zich voordoen bij de interpretatie van zulke gegevens ten behoeve van het beleid. De presentaties door vertegenwoordigers van overheid, contractlaboratoria en universiteiten, betroffen: toepassing van fysisch-chemische en ecotoxicologische testen voor de risico-beoordeling van slecht oplosbare stoffen ; het gebruik van aquatische toxiciteitsgegevens ten behoeve van de classificatie en risicobeoordeling ; de oplosbaarheid van 'moeilijke' stoffen ; de biologische beschikbaarheid van slecht oplosbare stoffen in aquatische toxiciteitsexperimenten ; praktische aspecten bij het vaststellen van de ecotoxiciteit van slecht oplosbare stoffen. Testen uitgevoerd met testconcentraties boven de wateroplosbaarheid of met een grote hoeveelheid oplosmiddel leveren resultaten die, gezien de biologische beschikbaarheid en het oplossingsproces, de werkelijke toxiciteit waarschijnlijk onderschatten. Voor risico-beoordelingsprocedures zijn testen met effecten boven de wateroplosbaarheidsgrens niet bruikbaar en kan om aanvullende testen worden gevraagd. Voor classificatie-doeleinden zou het afkeuren van zulke testen echter leiden tot de ongewenste situatie dat de stof niet wordt geclassificeerd. Voorgesteld wordt om in deze situaties de effecten van de stof te classificeren op het niveau van de oplosbaarheidsgrens van de stof. Geconcludeerd werd dat, gezien de grote hoeveelheid slecht oplosbare stoffen die voor regelgeving wordt aangemeld, aanvullende richtlijnen voor het uitvoeren van toxiciteitstesten (zoals de bereiding en analyse van testconcentraties) en voor de interpretatie van de testresultaten (zoals effectniveau's boven de oplosbaarheidsgrens) gewenst zijn.
dc.description.abstractThis document contains the papers, discussions and recommendations of a one-day seminar "Performing aquatic toxicity tests with poorly soluble substances" held in Bilthoven, the Netherlands on March 17, 1995. The great interest and participation in the workshop demonstrated the urgent need for more guidelines and harmonization among regulatory authorities and contract laboratories. This is necessary to solve the problems that occur when poorly soluble substances have to be tested and results have to be interpreted for classification and risk assessment procedures. The seminar was aimed at the exchange of information on performance and technical aspects, as well as on problems of policy interpretation relating to the ecotoxicological testing of organic compounds particularly difficult to dissolve in water. Subjects of the presentations held by government representatives, contract laboratories and universities were: risk assessment, substance classification, theory of solubility, bioavailability and practical aspects in ecotoxicity testing. Concluded was that tests performed with test concentrations above the solubility level or with a large amount of solvent probably result in an underestimation of true toxicity levels due to bioavailability and the solubility process. In risk assessment, tests with effect levels above the solubility level are not valid and further testing may be requested. Rejection of data for classification in these cases would mean no classification, which is undesirable. Therefore it was suggested to classify the effects at the water solubility level of the compound. Due to the large number of poorly soluble compounds submitted for registration it was concluded that additional guidelines are needed for performing toxicity tests (like the preparation and analysis of test concentrations) and for interpreting test results like effect levels above solubility limits.
