dc.description.abstractDe toxische effecten van zink voor de compostworm Eisenia andrei werden bestudeerd in twee proefveldexperimenten, om de veldrelevantie van laboratorium-toxiciteitsgegevens te onderzoeken indien blootstellingscondities op natuurlijke wijze fluctueren. Coconproductie werd hierbij gebruikt als sublethale toxiciteitsparameter. In 1995 werden waarnemingen gedaan na 7 en 8 weken blootstelling, en in 1996 na 3 en 7 weken. Waargenomen werd dat vocht en temperatuurcondities fluctueerden, en dat de blootstellingscondities afweken van de ecologische optima van de soort. Bovendien verschilden de abiotische condities tussen beide experimenten. In het experiment van 1995 waren de condities het minst optimaal, namelijk relatief droog, aan het begin van het experiment relatief koud, en aan het eind relatief warm. Dit kwam tot uiting in de lage coconproductie in schone proefveldgrond. Concentratie-effectrelaties werden berekend tussen enerzijds externe of interne (lichaams-) concentraties van zink en anderzijds de aantallen cocons. De EC50s die gebaseerd zijn op de lichaamsconcentraties van zink of op de wateroplosbare zinkconcentratie in de grond verschilden nauwelijks tussen de experimenten en de verschillende blootstellingsperioden. Dit suggereert dat deze concentraties nauw gerelateerd zijn aan de biobeschikbare zinkconcentratie, en dat de zinkgevoeligheid van E. andrei nauwelijks beinvloed wordt door blootstellingscondities. De EC50s die gebaseerd zijn op totale- en CaCl2 uitwisselbare externe zinkconcentraties waren voor 1996 hoger dan voor 1995. Dit werd veroorzaakt door een afname van de beschikbaarheid van zink door uitloging en toegenomen sorptie. De verhoogde EC50-waarden van 1996 duiden niet op een werkelijke verandering in de gevoeligheid van de dieren. Vergelijking van de proefveld-EC50 waarden met literatuurgegevens over zinkgevoeligheid onder gestandaardiseerde laboratoriumcondities bevestigt de conclusie dat de lichaamsconcentraties van zink en de wateroplosbare concentratie van zink in de grond nauw gerelateerd zijn aan de biobeschikbare concentratie. Het gebruik van deze concentratie-maten betekent een verlaging van de onzekerheden die bestaan bij laboratorium-veld extrapolatie van toxiciteitsgegevens bij deze soort.<br>

Cocoon production was used as the sublethal toxicity endpoint. In 1995, cocoon production was determined after 7 and 8 weeks of exposure, in 1996 after 3 and 7 weeks. Exposure conditions (temperature, humidity) fluctuated within the experiments. They also deviated from ecological optima derived from the literature and differed between the experiments. The harshest exposure conditions were present in 1995; in comparison to the optima, the conditions were too dry, initially too cold and eventually too hot. This was substantiated by a low cocoon production in control soil. Concentration-effect relationships between external (soil) or internal (body) zinc concentrations and cocoon production were calculated for both experiments and exposure durations. The EC50s based on body concentrations or on water-soluble soil concentrations of zinc were remarkably similar between experiments and exposure periods. This suggests that these concentration measures are closely associated to the bioavailable zinc fraction, and that zinc sensitivity of E. andrei is insensitive to exposure conditions. EC50s based on total and CaCl2 exchangeable zinc concentrations in the soil were highest for the 1996 experiment. This was caused by a decreased availability of zinc in the substrate due to leaching and increased sorption; the increase should not be interpreted as a change of true sensitivity of the animals. Comparison of the field plot results with zinc sensitivity of E. andrei under standardised laboratory test conditions confirmed the finding that body concentrations and water-soluble soil concentratrations of zinc are closely related to the bioavailable fraction. It is concluded that the use of these concentrations can be concluded to reduces uncertainties in laboratory-to-field extrapolation of toxicity data in this species.<br>
