dc.description.abstractHet is gebleken dat toediening van lage doses nitriet aan de rat gedurende 90 dagen hypertrofie van de zona glomerulosa van de bijnieren veroorzaakt. In bloed wordt nitriet snel omgezet in nitraat. Nitraat zou dus verantwoordelijk kunnen zijn voor deze afwijking in de bijnieren. Daarom werd in deze studie een vergelijking gemaakt van het effect van nitriet en nitraat op de zona glomerulosa. Drie groepen mannelijke Wistar ratten (elk bestaande uit 10 ratten) werden gedurende 90 dagen via het drinkwater blootgesteld aan respectievelijk kaliumchloride (controlegroep), kaliumnitriet en kaliumnitraat. De concentratie was steeds 36 mmol/l. De toename van het lichaamsgewicht van de ratten die aan nitriet of nitraat waren blootgesteld bleef iets achter bij de toename van het lichaamsgewicht van de controlegroep. De voeropname per kg lichaamsgewicht vertoonde geen verschillen tussen de drie behandelingsgroepen. De vloeistofconsumptie per kg lichaamsgewicht van de nitrietgroep was statistisch significant lager dan die van de controlegroep en van de nitraatgroep. Gedurende de eerste maand, maar niet daarna, waren de ratten van de nitrietgroep cyanotisch. Het voorbijgaand karakter van deze cyanose is mogelijk veroorzaakt door de grotere vloeistofinname (en bijgevolg ook een hogere belasting met nitriet) per kg lichaamsgewicht gedurende de eerste weken in vergelijking met de vloeistofinname gedurende de laatste weken. Aan het eind van de observatieperiode waren de methaemoglobineconcentratie in bloed en de nitrietconcentratie in plasma van de nitrietgroep statistisch significant hoger dan in de chloridegroep en in de nitraatgroep. De nitraatconcentraties in plasma van de nitriet- en nitraatgroep vertoonden onderling geen verschil maar waren duidelijk verhoogd ten opzichte van de controlegroep. Behandeling met nitriet of nitraat veroorzaakte geen eenduidig effect op de concentraties van T4, vrij T4, TSH, ACTH, corticosteron, en aldosteron in bloed. Bij de microscopische beoordeling werd in alle ratten van de nitrietgroep milde hypertrofie van de zona glomerulosa van de bijnieren waargenomen. In de ratten van de nitraatgroep daarentegen werd slechts incidenteel (2/10) minimale hypertrofie van de zona glomerulosa gezien. De morfometrische analyse van de bijnieren bevestigden de resultaten van de microscopishe beoordeling. De fractie van het oppervlak van de zona glomerulosa ten opzichte van de gehele bijnier in een mediane doorsnede van de bijnier was in de nitrietgroep statistisch significant groter dan die bij de controle- en nitraatgroep. De minimale hypertrofie van de zona glomerulosa die af en toe door de microscoop werd waargenomen bij de ratten die aan nitraat werden blootgesteld kon nauwelijks worden gedetecteerd door middel van morfometrie. Geconludeerd kan worden dat het nitraat ion geen rol speelt in het ontstaansproces van de hypertrofie van de zona glomerulosa van de bijnieren van de rat.<br>
dc.description.abstractFollowing administration of low doses of nitrite during 90 days, hypertrophy of the zona glomerulosa of the adrenals of rats has been reported. Nitrite in blood is readily converted into nitrate. Nitrate could have been responsible for this adrenal lesion. Therefore, the effect of subchronic administration of nitrite and nitrate on the adrenal zona glomerulosa were compared in this study. Three groups of 10 male Wistar rats each were exposed during 90 days to potassium chloride (control group), potassium nitrite, and potassium nitrate respectively by way of the drinking- water at a concentration of 36 mmol/l. The body weight gain of the rats exposed to nitrite and nitrate was slightly retarded as compared to the chloride dosed rats. No differences in the food intake per kg body weight were observed between the three treatment groups. The liquid consumption per kg body weight of the nitrite group was statistically significantly decreased as compared to the control and nitrate group. During the first month the rats receiving nitrite appeared cyanotic. This cyanotic appearance was not observed during the last two months. The transient character of this cyanotic appearance may have been caused by the higher intake of liquid (and consequently of nitrite) on a per kg body weight basis during the first weeks as compared to the liquid intake during the last weeks. At the end of the observation period, the methaemoglobin concentration in blood and the nitrite concentration in plasma of the nitrite group were statistically significantly increased as compared to the chloride and nitrate groups. The nitrate concentrations in plasma of the nitrite and nitrate groups did not differ between each other but they were increased as compared to the control group. Treatment with nitrite or nitrate induced no consistent effect on the concentrations of T4, free T4, TSH, ACTH, corticosteron, and aldosteron in blood. On microscopic examination, slight hypertrophy of the adrenal zona glomerulosa was observed in all nitrite dosed rats while in the rats exposed to nitrate, minimal hypertrophy of the adrenal zona glomerulosa was found occasionally (2/10). The results of the morphometric analysis of the adrenals were in line with the microscopical examination of the adrenals. In the nitrite dosed rats the fraction of the surface area of zona glomerulosa in a median section of the adrenal gland, was statistically significantly increased as compared to the control and nitrate groups. The minimal hypertrophy of the adrenal zona glomerulosa observed occasionally on microscopic examination in the rats exposed to nitrate could hardly be detected by morphometric analysis. It was concluded that nitrate ion does not play a role in the etiology of the hypertrophy of the zona glomerulosa of the adrenal glands of the rat.<br>
