dc.description.abstractEen on-line methode is ontwikkeld voor een gecombineerde derivatisering en analyse met behulp van LC-ESI/MS van peptides op femto-mol niveau. Peptides werden na belading op een trapkolom in een microkolom schakelsysteem gederivatiseerd met N-succinimidyl-2(3-pyridyl)acetate (SPA) in gebufferd aqueous medium by pH 7. Het gemakkelijk hydrolyseerbare reagens was opgelost in droge methanol en werd vervolgens in het HPLC systeem gemengd in een 3% volume ratio met de buffer en vervolgens over het monster op de trapkolom geleid. Na de reactie en het wassen van de voorkolom met water werden de reactieproducten (derivaten) met behulp van een gradientelutie van de trapkolom overgebracht naar de analytische kolom voor scheiding en ESI tandam MS analyse. Een vrijwel kwantitatieve reactie vond plaats met N-terminale en lysine amino groepen. De reactie met de lysine aminogroep verschaft een doeltreffende chemische methode voor differentiatie tussen de isobare residuen glutamine en lysine. In een aantal gevallen vond een geringe reactie plaats met de phenolische OH groep van tyrosine. De pyridylacetyl groep op de N-terminale aminogroep kan leiden tot een verandering van het fragmentatiepatroon in MS/MS ten gunste van de vorming van zogenaamde, hetgeen aanvullende informatie levert in MS/MS spectra in sequentie-analyses.<br>

The fast hydrolysing reagent was solved in dry methanol and mixed in a 3 vol% ratio with a buffer just before reaching the sample on the PC. Following the reaction and wash, the N-pyridylacetyl (PA) derivatives formed were transferred to the analytical micro-HPLC column for separation and subsequent ESI tandem MS analysis. The reaction was nearly quantitative and takes selectively place at the N-terminal and lysine amino functions, the latter providing a chemical means to distinguish between isobaric residues lysine (K) and glutamine (Q) in peptide sequencing. In some cases, a minor reaction was observed with the tyrosine hydroxyl group. The N-terminal PA groups was capable to alter the collision induced fragmentation (CID) pathway of peptides in favour to the formation of abundant b-type ions, a helpful feature for sequence elucidation of unknown peptides, particularly for quadrupole ion trap instruments.<br>
