dc.description.abstractQuantitative Structure Activity Relationship (QSAR) modelling techniques are overviewed here, along with descriptors which can be used in QSAR equations and the different statistical methods suitable for deriving QSARs. Discussed is the current state of the art on the use of QSAR estimates within the different regulatory assessment frameworks of the Centre for Substances and Risk Assessment at the Dutch National Institute of Public Health and the Environment (RIVM/CSR). Special emphasis has been placed on environmental effect assesssments. Commonly accepted QSAR equations and software programs used in environmental exposure assessments are described. Generally accepted QSAR equations and other QSARs for environmental effects documented in the literature are presented in tabular form; compounds acting by non-polar narcosis, polar narcosis, reactive chemicals and chemicals with a specific mode of action are presented separately. Recently developed computerised QSAR programs are also overviewed. Several recommendations are made for the use of QSARs within RIVM/CSR risk/hazard assessment frameworks, with experiences related to routine application of QSARs in effect assessments to be evaluated and reported on within one year.
dc.description.abstractEen overzicht is gegeven van modelleringstechnieken voor Kwalitatieve Stuctuur Activiteit Relaties (QSAR) evenals van descriptoren die kunnen worden gebruikt bij QSAR berekeningen en van verschillende statistische methoden, die geschikt worden geacht voor de afleiding van QSAR's. De huidige stand van zaken ten aanzien van het gebruik van QSAR's binnen de verschillende reguliere beoordelingskaders van het Centrum voor Stoffen en risicobeoordeling van het Nationaal Instituut voor Volksgezondheid en Milieu (RIVM/CSR) wordt toegelicht. De algemeen geaccepteerde QSAR berekeningen en software programma's voor de beoordeling van ecotoxicologische risico's via de blootstellingsroute zijn beschreven. De nadruk ligt echter op de beoordeling van ecotoxicologische effecten. Algemeen geaccepteerde QSAR's en andere in de openbare literatuur vastgelegde QSAR's voor de beoordeling van ecotoxicologische effecten worden in aparte tabellen gepresenteerd voor respectievelijk stoffen die werken via niet-polaire narcose, polaire narcose, reactieve stoffen en stoffen met een specifiek werkingsmechanisme. Ook wordt een overzicht gegeven van recentelijk ontwikkelde QSAR computer programma's. Enkele aanbevelingen worden gedaan op welke wijze QSAR's binnen de beoordelingskaders van RIVM/CSR kunnen worden toegepast. Ervaringen met routinematig toepassen van QSAR's bij de beoordelingen van ecotoxicologische effecten worden over een jaar geevalueerd en gerapporteerd.
